Short description :
DNA viruses detection (WSSV, IHHNV, BP, MBV y HPV) using nested-PCR technique is based in a double set of cycles amplified (carried out) by a Taq DNA polymerase, using a pair of specific primers from a genomic region of the virus for its detection in shrimp hemocytes, larvae and tissues. Amplified products are analyzed in electrophoresis gel of agarose a 2% stained with ethidium bromide, through ultra-violet light observation (UV).

Analysis using RT-nested PCR for the detection of RNA viruses (IMNV, TSV, YHV, GAV, LSNV).

The detection of IMNV, TSV, YHV, GAV and LSNV viruses using he RT-PCR method is based on an inverse transcriptase that converts viral RNA into Complementary DNA, which is used as a target for the amplification reaction “carried out” by the DNA Taq polymerase, using two pairs of specific primers from a genomic region of the virus, for its detection in shrimp hemocytes, larvae and tissues. Amplified products are analyzed in an electrophoresis gel of agarose at 2% stained with ethidium bromide through its observation with ultra-violet light (UV).

La detección de los virus IMNV, TSV, YHV, GAV y LSNV mediante el método de RT-PCR (PCR con transcripción inversa) se basa en la acción de una Transcriptasa inversa que convierte el ARN viral en ADN complementario, el cual sirve de blanco para la reacción de amplificación mediada por la Taq ADN polimerasa, utilizando cebadores específicos de una región genómica de los virus, para su detección en hemocitos, larvas y tejidos del camarón. Los productos amplificados son analizados en un gel de electroforesis de agarosa al 2% teñido con bromuro de etidio, mediante la observación con luz ultravioleta (UV). |
